Cholesterol-induced fluid membrane domains: a compendium of lipid-raft ternary phase diagrams.

چکیده
The biophysical underpinning of the lipid-raft concept in cellular membranes is the liquid-ordered phase that is induced by moderately high concentrations of cholesterol. Although the crucial feature is the coexistence of phase-separated fluid domains, direct evidence for this in mixtures of cholesterol with a single lipid is extremely sparse. More extensive evidence comes from ternary mixtures of a high chain-melting lipid and a low chain-melting lipid with cholesterol, including those containing sphingomyelin that are taken to be a raft paradigm. There is, however, not complete agreement between the various phase diagrams and their interpretation. In this review, the different ternary phase diagrams of cholesterol-containing systems are presented in a uniform way, using simple x,y-coordinates to increase accessibility for the non-specialist. It is then possible to appreciate the common features and examine critically the discrepancies and hence what direct biophysical evidence there is that supports the raft concept.

Observation of Fluid–Fluid Immiscibility in Ternary Mixtures of DPPC, DOPC and Cholesterol
As discussed chapter 1, cholesterol–rich domains in biomembranes, called rafts, are believed to be involved in many cellular functions [1, 2, 3, 4, 5]. Biochemical studies on the insoluble membrane fraction obtained from detergent extraction of plasma membranes show that it mainly consists of saturated lipids, such as sphingomyelin and cholesterol. The detergent insoluble fraction, known as det...
متن کاملLiquid-ordered phases induced by cholesterol: a compendium of binary phase diagrams.
Mixtures of phospholipids with cholesterol are able to form liquid-ordered phases that are characterised by short-range orientational order and long-range translational disorder. These L(o)-phases are distinct from the liquid-disordered, fluid L(alpha)-phases and the solid-ordered, gel L(beta)-phases that are assumed by the phospholipids alone. The liquid-ordered phase can produce spatially sep...
متن کاملProbing lipid mobility of raft-exhibiting model membranes by fluorescence correlation spectroscopy.
Confocal fluorescence microscopy and fluorescence correlation spectroscopy (FCS) have been employed to investigate the lipid spatial and dynamic organization in giant unilamellar vesicles (GUVs) prepared from ternary mixtures of dioleoyl-phosphatidylcholine/sphingomyelin/cholesterol. For a certain range of cholesterol concentration, formation of domains with raft-like properties was observed. S...
متن کاملCoarse-grained simulations of branched bilayer membranes: effects of cholesterol-dependent phase separation on curvature-driven lipid sorting
Our recent coarse-grained (CG) molecular dynamics (MD) simulations of membranes with a hemifused-ribbon (λ-shaped) geometry showed curvature-driven demixing leading to enrichment in dioleoyl-phosphatidylethanolamine (DOPE) in a negatively-curved region (at C –0.8 nm) of a DOPE/dipalmitoyl-phosphatidylcholine (DPPC) membrane. Here we extend the analysis with respect to lipid composition and simu...
متن کاملCorrelated AFM and NanoSIMS imaging to probe cholesterol-induced changes in phase behavior and non-ideal mixing in ternary lipid membranes.
Cholesterol is believed to be an important component in compositionally distinct lipid domains in the cellular plasma membrane, which are referred to as lipid rafts. Insight into how cholesterol influences the interactions that contribute to plasma membrane organization can be acquired from model lipid membranes. Here we characterize the lipid mixing and phase behavior exhibited by (15)N-dilaur...
